How did users rate their sleep, physical function and other aspects of everyday health over six weeks? This article brings together paired questionnaires from 20 NovaStemix® users, presents the results across all six domains, and explains some of the scientific background to the ingredient choices.

Among the 20 users who completed follow-up and met the inclusion criteria, group mean scores moved in a favourable direction across all six domains. General health, physical function, sleep disturbance and self-reported cognitive function remained statistically significant after adjustment for multiple comparisons. Fatigue and pain interference did not meet this threshold. This observation describes changes over time; it does not establish that NovaStemix caused them.

Why we recorded everyday experiences

Sleep, ease of daily activity and tiredness are matters that users care about. Recording these experiences with the same questionnaires at two time points helps turn individual feedback into information that can be compared.

Questionnaires capture participants' own assessments of their health and daily functioning. We compared each user's scores before starting NovaStemix with their scores at week six. This describes changes during that period. Understanding how much, if any, was attributable to the product requires consideration of other influences.

Whose data were included

Forty questionnaires were distributed. The final analysis included complete paired data from 20 users, comprising nine men and eleven women.

Twenty other records were not included, mainly because of irregular or discontinued use, a missing week-six questionnaire, or intake below two capsules per day. Separate counts for each exclusion reason were not available. The results describe the 20 users included in the analysis, rather than everyone initially involved or everyone who purchases the product.

The recommended intake is four capsules daily. Actual intake among those included ranged from two to four capsules daily. This was follow-up of everyday use, with variation in intake. These data are insufficient to compare different doses. Routine use should follow the product label.

Questionnaires and analysis

Participants completed the relevant PROMIS questionnaires before starting use, labelled W0, and at week six, labelled W6. The six domains were general health, fatigue, physical function, sleep disturbance, pain interference and self-reported cognitive function.

In the general health questionnaire, one pain-intensity item that had caused differences in interpretation was removed. The remaining nine items were combined into a custom total. This is not a standard PROMIS Global Health score, and no official Global Health T-score was assigned to it. The separate six-item pain interference questionnaire was retained.

Each domain had 20 complete pairs. We analysed the within-person difference, W6 minus W0, using two-sided paired t-tests. The Holm method adjusted for the six comparisons. Statistical significance was assessed using an adjusted p value below 0.05.

Higher scores indicate a favourable direction for general health, physical function and cognitive function. Lower scores indicate a favourable direction for fatigue, sleep disturbance and pain interference. The domains differ in their items, scoring and meaning, so comparisons are made within each domain over time.

General health, physical function, sleep disturbance and cognitive function met the adjusted significance threshold. Mean fatigue and pain interference scores also moved in a favourable direction, but did not meet that threshold.

As a secondary check, the five complete short forms were converted to PROMIS T-scores for each participant at each time point using the corresponding official conversion tables. The five T-score comparisons and the modified general health raw-score comparison were then considered together with Holm adjustment across six tests. The pattern of statistical significance was unchanged.

How to interpret the findings

Lower sleep disturbance scores mean that, on average, the users included reported less sleep-related difficulty at week six than before starting use. Sleep monitoring was not used to verify changes in sleep duration or sleep architecture.

Higher physical function and general health scores reflect favourable changes in participants' assessments of daily activity and overall health. The general health result must be interpreted in the context of the modified nine-item scoring used here.

Higher cognitive function scores reflect changes in how participants perceived their cognitive functioning. They cannot be translated directly into improvements in objective memory or cognitive test performance.

Fatigue and pain interference did not reach statistical significance. This dataset therefore provides insufficient statistical evidence of a group-level change in these two domains. Their lower mean scores should be reported together with this uncertainty.

These findings describe experiences reported during six weeks of NovaStemix use. Recording everyday experiences has value, but statistical significance does not establish clinical importance or imply that every user will have the same experience.

Scientific context for the ingredient choices

The thinking behind the formula draws on research into cellular energy metabolism, recycling of damaged cellular components and responses to stress. A few examples illustrate this background.

Cellular energy metabolism and nicotinamide riboside. Nicotinamide riboside, or NR, is a precursor used to make NAD+, which participates in cellular energy metabolism. A placebo-controlled human trial reported higher NAD+ concentrations in blood cells after NR supplementation. [1]

Cellular recycling and spermidine. Autophagy is a process through which cells break down and recycle cellular material, including damaged components. Animal research on spermidine has examined autophagy and mitochondrial maintenance, offering clues about how cells maintain their function. [2]

Cellular stress responses and plant compounds. Research on curcumin and sulforaphane-related compounds has examined defence pathways regulated by Nrf2. A small randomised human study of glucoraphanin, curcumin or their combination reported changes in certain protective and inflammation-related gene-expression measures in skin samples. Those findings concern the tissues and markers measured. [3]

These studies help explain our interest in potentially complementary ingredient roles. Connecting these mechanisms with the questionnaire changes in daily function or sleep is a hypothesis that could be tested further. This follow-up did not measure NAD+, autophagy or inflammatory markers. It cannot confirm whether those pathways changed or attribute a questionnaire change to a particular ingredient.

The doses, formulations and populations in ingredient studies may differ from those relevant to NovaStemix. Such studies do not establish that the finished NovaStemix formula has a synergistic effect.

Scope and limitations

The sample was small, with no randomisation or control group, and intake varied. Lifestyle changes, concurrent treatment, other supplements, natural fluctuations and participants' expectations could all have influenced the responses. Users who completed follow-up may also differ from those whose records were not included. The findings therefore support a description of changes reported by the included users, rather than a conclusion about NovaStemix's independent effect.

The raw questionnaire scales are not proportional measures of health. A percentage change in these scores cannot be interpreted as a percentage improvement in health or as a product success rate.

We have made these completed customer follow-up data available so that users and partners can examine the findings, methods and limits of interpretation.

Statistical details

The following paired tests use raw total scores, with 19 degrees of freedom in each t-test. Cohen's dz is the mean paired difference divided by the standard deviation of the paired differences. It describes the size of the within-group change. A negative sign indicates a lower score; the favourable direction depends on the domain, as explained above.
